The point-mutated feline HER2 DNA vaccine (also known as pfeHER2-K or feHER2-K) is a research-stage veterinary DNA vaccine designed to overcome immune tolerance to the HER2 self-antigen in cats. It consists of a plasmid DNA construct encoding a feline HER2 protein with a single amino acid point mutation (specifically the Q141K substitution), which introduces a positive charge on an exposed surface region. This modification renders the self-antigen sufficiently foreign to break immune tolerance and stimulate both humoral and cellular immune responses while preserving the overall HER2 protein structure and epitopes. Administered via intramuscular electrovaccination (DNA electroporation), the vaccine is being investigated for the treatment of feline mammary carcinoma (FMC), a spontaneous cancer in domestic cats that closely mirrors HER2-positive human breast cancer.
