Overview

POL7306 is a first-in-class, synthetic peptidomimetic outer-membrane-protein-targeting antibiotic (OMPTA) being developed by Spexis (formerly Polyphor) as a medium-spectrum agent against multidrug-resistant Gram-negative bacteria, including WHO priority 1 and ESKAPE pathogens. It binds to lipopolysaccharides (LPS) and the essential outer membrane protein BamA, disrupting outer membrane biogenesis and integrity, leading to rapid bactericidal activity and cell lysis.[1][3][7][9] Derived from the company’s Protein Epitope Mimetics platform, POL7306 has shown potent in vitro and in vivo activity against colistin-resistant, extensively drug-resistant, and carbapenemase-producing clinical isolates, with a low propensity for resistance development, and is in preclinical/early clinical development for serious Gram‑negative infections.[1][4][5][6][9][10]
