The POLA1 inhibitor is a preclinical-stage small molecule therapeutic being developed by Cumulus Oncology in collaboration with the Lead Discovery Center (LDC) in Germany. It targets the DNA polymerase alpha catalytic subunit (POLA1), which is the enzyme responsible for the initiation of DNA replication by synthesizing RNA-DNA primers on both the leading and lagging strands. By inhibiting POLA1, the drug interferes with the DNA replication process, leading to replication stress and potential cell death in rapidly dividing cancer cells. The program is currently in the discovery to lead optimization phase, focusing on oncology indications with high unmet medical needs.
