The polar methanolic fraction (PMF) is a natural product extract derived from *Hypericum perforatum* L. (St. John's Wort) that contains high concentrations of photoactive naphthodianthrones, primarily hypericin and pseudohypericin. It is developed as a natural photosensitizer for use in photodynamic therapy (PDT) and photodynamic diagnosis (PDD). When exposed to specific wavelengths of light, typically around 630 nm, PMF generates reactive oxygen species (ROS) and singlet oxygen, which induce selective oxidative stress and trigger apoptosis in malignant cells. Preclinical studies have demonstrated that PMF exhibits significant photocytotoxicity against human bladder cancer cell lines (including metastatic T24 and low-grade RT4) and leukemic cells (HL-60), often showing efficacy comparable to or exceeding that of clinically approved synthetic photosensitizers like Photofrin.
