Overview
Polatuzumab vedotin + rituximab + dexamethasone + cytarabine + cisplatin is a multi-agent chemotherapy and immunotherapy regimen under investigation for the treatment of relapsed or refractory diffuse large B-cell lymphoma (DLBCL) and other aggressive B-cell lymphomas. Polatuzumab vedotin is an antibody-drug conjugate targeting CD79b on B cells; it delivers the microtubule inhibitor monomethyl auristatin E (MMAE) to malignant cells[1][2]. Rituximab is a monoclonal antibody against CD20 on B lymphocytes. Dexamethasone is a synthetic glucocorticoid with anti-inflammatory and immunosuppressive effects. Cytarabine is an antimetabolite that inhibits DNA synthesis. Cisplatin is a platinum-based chemotherapeutic agent that crosslinks DNA to induce apoptosis in rapidly dividing cells. This combination aims to maximize antitumor activity by leveraging multiple mechanisms of action.
