Polatuzumab vedotin + rituximab + gemcitabine + dexamethasone + cisplatin (PV-RGDP) is an investigational chemoimmunotherapy combination regimen being evaluated for the treatment of relapsed or refractory diffuse large B-cell lymphoma (rrDLBCL). The regimen integrates polatuzumab vedotin, an antibody-drug conjugate (ADC) targeting CD79b, with the standard R-GDP salvage chemotherapy backbone. Polatuzumab vedotin delivers the potent microtubule inhibitor monomethyl auristatin E (MMAE) directly to malignant B-cells, while rituximab provides anti-CD20 mediated immunotherapy. The chemotherapy components—gemcitabine, cisplatin, and dexamethasone—provide synergistic cytotoxic effects through DNA damage and anti-proliferative mechanisms. This specific combination is currently being studied in a Phase II clinical trial led by the UNC Lineberger Comprehensive Cancer Center to improve salvage therapy outcomes for both transplant-eligible and transplant-ineligible patients.
