Overview

Pola-R-ICE is an experimental chemoimmunotherapy combination regimen being evaluated as a salvage therapy for patients with relapsed or refractory diffuse large B-cell lymphoma (DLBCL) who are candidates for autologous stem cell transplant (ASCT). The regimen consists of the antibody-drug conjugate polatuzumab vedotin added to the standard-of-care R-ICE (rituximab, ifosfamide, carboplatin, and etoposide) backbone. Polatuzumab vedotin targets CD79b, a component of the B-cell receptor, delivering the cytotoxic payload monomethyl auristatin E (MMAE) directly to malignant B cells. This combination aims to improve complete response rates and progression-free survival compared to R-ICE alone in the second-line setting. The Phase III trial (NCT04662255) investigating this regimen is sponsored by GWT-TUD GmbH.
