Overview
AD + CP is a chemotherapy-immunotherapy combination regimen being evaluated in the RADICAL clinical trial (NCT05253495) sponsored by New York Medical College. The regimen consists of the antibody-drug conjugate polatuzumab vedotin (Polivy), the anthracycline doxorubicin (Adriamycin), the alkylating agent cyclophosphamide, and the corticosteroid dexamethasone. It is specifically being investigated for the treatment of children, adolescents, and young adults with newly diagnosed mature B-cell non-Hodgkin lymphoma (MB-NHL) or classical Hodgkin lymphoma (cHL). The study aims to assess whether adding targeted immunotherapy can allow for a reduction in anthracycline exposure and radiation therapy while maintaining disease control, thereby minimizing long-term toxicities. Polatuzumab vedotin targets CD79b, a protein expressed on the surface of B cells, and delivers the cytotoxic microtubule inhibitor monomethyl auristatin E (MMAE) directly to malignant cells.
