Overview

POLEP286R neoantigen peptide is a tumor-specific neoantigen derived from a recurrent hotspot mutation (P286R) in the DNA polymerase epsilon (POLE) gene, which is frequently found in a subset of endometrial cancers characterized by an ultramutated phenotype. This peptide is specifically presented on the cell surface by HLA-A11:01 molecules. As a neoantigen, it is recognized as "non-self" by the immune system, making it a highly specific target for T-cell-based immunotherapies, such as peptide vaccines or TCR-engineered T-cell therapies. Research is currently focused on identifying high-affinity T-cell receptors (TCRs) that can recognize this peptide-HLA complex to induce a potent anti-tumor immune response in patients with POLE-mutant endometrial cancer.
