Overview

Salk IPV is a trivalent inactivated poliovirus vaccine (IPV) derived from wild-type poliovirus strains: Type 1 (Mahoney), Type 2 (MEF-1), and Type 3 (Saukett). Originally developed by Jonas Salk in the 1950s, this vaccine is produced by growing the viruses in Vero cell cultures or monkey kidney cells, followed by purification and inactivation with formaldehyde. It is the global standard for inactivated polio vaccination, providing high levels of serum neutralizing antibodies that protect against paralytic poliomyelitis. Unlike the live-attenuated oral polio vaccine (OPV), Salk IPV contains no live virus and therefore cannot cause vaccine-associated paralytic poliomyelitis (VAPP) or lead to the emergence of vaccine-derived polioviruses (VDPVs). It is typically administered via intramuscular injection and is frequently used as the reference standard in clinical trials for next-generation Sabin-strain IPVs (sIPV).
