Drug intelligence / Profile preview
POLK-38 is an orally active, brain-penetrant small molecule modulator of prolyl oligopeptidase (PREP) developed by Polku Therapeutics, a spinout from the University of Helsinki. It targets the PREP–PP2A (protein phosphatase 2A) pathway to restore protein homeostasis and promote tau dephosphorylation. In preclinical models of tauopathy, such as the PS19 mouse model, oral administration of POLK-38 has demonstrated significant efficacy in reducing brain tau pathology and improving spatial learning. It is being developed as a disease-modifying therapy for neurodegenerative disorders, including Alzheimer's disease, frontotemporal dementia, and traumatic brain injury.
