Overview

Poloxamer 407 is a hydrophilic non-ionic surfactant and triblock copolymer composed of a central hydrophobic block of polypropylene glycol flanked by two hydrophilic polyethylene glycol blocks. It is primarily used as an excipient in pharmaceutical formulations for its surfactant, emulsifying, solubilizing, dispersing, and absorbance-enhancing properties. Poloxamer 407 exhibits thermoreversible gelation—remaining fluid at room temperature but forming a gel at body temperature—which makes it valuable for controlled drug delivery applications across oral, rectal, topical, ophthalmic, nasal and injectable preparations. It is also widely used in cosmetics (e.g., contact lens solutions), mouthwashes and bioprinting due to its unique phase-change properties. While generally considered pharmacologically inactive as an excipient (not acting on specific biological targets), research has explored its potential effects on lipid profiles and renal function when administered parenterally[1][2][4][5].
