Drug intelligence / Profile preview
POLR2A siRNA is a small interfering RNA (siRNA) designed to target and silence the expression of the POLR2A gene, which encodes RNA polymerase II subunit A (RPB1), the largest subunit of the RNA polymerase II complex. In preclinical research, POLR2A siRNA has been investigated for its therapeutic potential in acute kidney injury (AKI) and chronic kidney disease (CKD) by preventing proximal tubule cells from being trapped in a dedifferentiated state. Additionally, POLR2A siRNA has been studied as a targeted therapy for cancers harboring hemizygous loss of TP53 and POLR2A (such as triple-negative breast cancer and colorectal cancer), exploiting a collateral vulnerability where partial loss of POLR2A renders cancer cells highly sensitive to further POLR2A suppression.
