Drug intelligence / Profile preview
PRTX007 + pembrolizumab is an investigational combination therapy being evaluated for the treatment of cancer, specifically resectable Stage III melanoma in the neoadjuvant setting. The combination consists of polvitolimod hydrochloride (PRTX007), an orally administered, systemically acting small-molecule prodrug that agonizes Toll-like receptor 7 (TLR7), and pembrolizumab (Keytruda), a humanized IgG4 monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor. Polvitolimod is designed to selectively activate the interferon regulatory factor 7 (IRF7) pathway to drive a poly-interferon response while avoiding the pro-inflammatory NF-κB pathway. By combining innate immune activation via TLR7 agonism with adaptive immune checkpoint inhibition via PD-1 blockade, the regimen aims to enhance anti-tumor immune responses and improve pathologic response rates prior to surgical resection.
