Poly(L-glutamic acid)-grafted hyaluronan (PGA-g-HA) is a synthetic hyaluronan derivative designed for intra-articular injection to treat osteoarthritis. By grafting poly(L-glutamic acid) onto a hyaluronan backbone, the molecule achieves significantly enhanced resistance to enzymatic degradation by hyaluronidase, addressing the short half-life of conventional hyaluronic acid viscosupplements. In preclinical studies, PGA-g-HA demonstrated viscoelastic properties similar to high-molecular-weight hyaluronan and exhibited potent anti-inflammatory activity by inhibiting IL-1β-mediated inflammatory responses in chondrocytes. It has shown superior chondroprotective effects in rat models of knee osteoarthritis compared to unmodified hyaluronan, suggesting its potential as a durable, disease-modifying treatment for joint degeneration.
