Overview

Poly ICLC is a synthetic immunostimulant composed of polyinosinic-polycytidylic acid (poly I:C) complexed with the stabilizers carboxymethylcellulose and poly-L-lysine. It acts as a viral double-stranded RNA mimic, recognized by pattern recognition receptors such as toll-like receptor 3 (TLR3), MDA5, and DDX/DHX RNA helicases. By activating these innate immune sensors, Poly ICLC induces the production of type I interferons and pro-inflammatory cytokines, enhances dendritic cell activation, promotes Th1 polarization, and stimulates cytotoxic T cell responses. Its primary clinical development has been as an adjuvant in cancer immunotherapy—both as a direct antitumor agent and to enhance vaccine efficacy—and it is also being investigated for other indications involving immune modulation[1][2][3][4][5][6].
