Poly-ICLC-matured dendritic cells are an investigational autologous cell-based immunotherapy. The therapeutic process involves harvesting a patient's monocytes, differentiating them into dendritic cells, and loading them with specific tumor-associated antigens (such as NY-ESO-1, Melan-A, hTERT, CEA, or autologous tumor lysate). These cells are then matured ex vivo using Poly-ICLC (Hiltonol), a stabilized double-stranded RNA analogue that acts as a potent agonist for Toll-like receptor 3 (TLR-3) and MDA-5. This maturation step is critical for enhancing the dendritic cells' ability to present antigens and secrete pro-inflammatory cytokines. Once re-administered to the patient, typically via intradermal injection, these matured dendritic cells activate cytotoxic T lymphocytes and natural killer (NK) cells to mount a targeted anti-tumor immune response. This platform has been evaluated in Phase I and Phase II clinical trials for various malignancies, including glioblastoma, melanoma, and pancreatic cancer.
