Overview

Polyadenylic-polyuridylic acid is a synthetic double-stranded RNA molecule composed of alternating strands of adenylic and uridylic acids. It functions as an immunostimulant by mimicking viral double-stranded RNA, thereby activating innate immune responses primarily through Toll-like receptor 3 (TLR3). This activation leads to TRIF-dependent signaling, resulting in the induction of pro-inflammatory cytokines and type I interferons via NF-kB and IRF pathways. The drug has demonstrated antitumor effects in preclinical models and was investigated as an adjuvant therapy for operable breast cancer, where it showed improved survival outcomes in certain patient subgroups. It also exhibits antiviral activity against viruses such as HIV in cell culture. Clinical studies have shown that it enhances natural killer (NK) cell activity and interferon-mediated protein kinase levels without significant toxicity at tested doses[1][2][4][5][6][8].
