Polyclonal Regulatory T Cells (PolyTregs) are a cell therapy consisting of expanded autologous regulatory T cells (Tregs). Tregs are a subpopulation of CD4+ T cells that modulate the immune system, maintain tolerance to self-antigens, and prevent autoimmune disease. They express biomarkers CD4, FOXP3, and CD25. The manufacturing process typically involves isolation of CD4+CD25+CD127lo/- Tregs from a patient's peripheral blood using fluorescence-activated cell sorting (FACS), followed by expansion for approximately 14 days in medium containing IL-2 and stimulation with anti-CD3/CD28 beads. Polyclonal Tregs are being investigated as a potential treatment for autoimmune diseases, solid organ transplantation, and graft versus host disease due to their immunosuppressive properties.
