Overview

Polydatin is a naturally occurring stilbenoid glucoside and the main glycosylated precursor of resveratrol. It is primarily isolated from the roots and rhizomes of *Polygonum cuspidatum* (also known as *Reynoutria japonica*) and can also be found in grapes, peanuts, hop cones, red wines, cocoa-containing products, and chocolate[1][2][5]. Polydatin exhibits a broad range of pharmacological activities including anti-inflammatory, immunoregulatory, antioxidant (notably superior to resveratrol), neuroprotective, cardioprotective, hepatoprotective and anti-tumor effects[1][2][5]. Mechanistically it modulates key signaling pathways involved in inflammation (such as NF-kB), oxidative stress (e.g., Nrf2/HO1 pathway), apoptosis regulation (e.g., Bcl2/Bax ratio), platelet aggregation inhibition and neuroprotection[8]. It has higher bioavailability than resveratrol due to its glycosylation. Traditionally used in Chinese medicine for analgesic, antipyretic and diuretic purposes[4], polydatin has entered clinical trials for conditions such as hemorrhagic shock and irritable bowel syndrome. Recent studies have explored its potential benefits in ischemic cerebrovascular disease/microvascular disease/shock/burns/traumatic shock (Phase 2)[8], neuropathic pain[5], bone/joint disorders[10], vascular dementia[3] and Alzheimer's disease models[7].
