Overview

Polyestradiol phosphate is a long-lasting estrogen medication primarily used in the treatment of prostate cancer in men. It is also used for breast cancer in women, as hormone therapy for low estrogen levels and menopausal symptoms, and as part of feminizing hormone therapy for transgender women. Administered by intramuscular injection every four weeks, it acts as a prodrug that slowly releases estradiol after metabolism. As an agonist of the estrogen receptor, it exerts its effects by mimicking endogenous estrogens. Polyestradiol phosphate is a polymeric ester of estradiol and phosphoric acid with an extended duration of action due to its slow release and conversion to active estradiol in the body. The drug was discovered around 1953 and introduced into medical use in 1957; it was initially developed by Pfizer but has since been withdrawn from several markets including the United States[1][2][5][8].
