Polyethylene glycol 400 (PEG 400) is a low-molecular-weight grade of polyethylene glycol, characterized by an average molecular weight of approximately 400. It is a clear, colorless, viscous liquid with strong hydrophilic properties and very low toxicity. PEG 400 is widely used in pharmaceutical formulations as an excipient to enhance the solubility and bioavailability of poorly water-soluble drugs. In addition to its role as a solvent, plasticizer, surfactant, ointment base, suppository base, and tablet/capsule lubricant in drug manufacturing, it serves as an active ingredient in ophthalmic solutions for the relief of dry eye symptoms by acting as a synthetic ocular lubricant (lacrimomimetic). Polyethylene glycols are also used for PEGylation—attaching them to protein medications to improve their solubility and pharmacokinetics[1][2][3][4][5].
