Overview

**Polyethylene glycol thymosin alpha1 (PEG-Tα1)** is a pegylated conjugate of thymosin alpha1, a 28-amino acid synthetic peptide derived from the thymus gland, designed as a long-acting immunomodulator to extend the half-life and pharmacokinetics of the parent peptide. It enhances immune function by stimulating dendritic cell maturation, promoting cytokine production such as IL-12, increasing T-cell proliferation and differentiation, restoring lymphokine-activated killer cell activity, and augmenting antigen presentation via Toll-like receptor signaling. Primarily investigated in combination with antivirals like adefovir for chronic hepatitis B, PEG-Tα1 has demonstrated efficacy in improving viral response rates and safety in phase III trials, positioning it as a category 1.1 chemical drug in certain regulatory contexts, with potential broader applications in viral infections due to its tolerability and immune-enhancing profile.[1][2][14]
