Polyethylenglycol-treated gammaglobulin is a PEGylated formulation of human normal immunoglobulin G (IgG), in which the immunoglobulins are chemically modified with polyethylene glycol (PEG) to enhance pharmacokinetic properties such as half-life and stability. This modification aims to reduce immunogenicity and renal clearance, allowing for less frequent dosing and potentially improved safety profiles. The drug is used primarily as replacement therapy in primary immunodeficiency disorders, for prevention of infections in B-cell chronic lymphocytic leukemia-associated hypogammaglobulinemia, treatment of idiopathic thrombocytopenic purpura (ITP), and prevention of coronary artery aneurysms in Kawasaki syndrome. Its mechanism involves providing passive immunity by supplying exogenous IgG antibodies that neutralize pathogens and modulate immune responses through interactions with cytokines, complement proteins, Fc receptors on immune cells, and autoantibodies[2][3][8][9].
