polyG3 is a synthetic, guanosine-rich oligonucleotide that functions as a selective agonist for Toll-like receptor 8 (TLR8). It is primarily utilized in immunological research to study the activation of the TLR8 pathway and its downstream effects on innate and adaptive immune responses. In oncology research, particularly concerning multiple myeloma, polyG3 is used to investigate the modulation of myeloid-derived suppressor cells (MDSCs). By activating TLR8, polyG3 can attenuate the MDSC-mediated induction of T-cell senescence, suggesting a potential mechanism for overcoming immune evasion in the tumor microenvironment. The agent has been shown to reduce SA-β-gal-positive staining in T cells when co-cultured with treated monocytic MDSCs (M-MDSCs), thereby potentially reversing the immunosuppressive effects that facilitate tumor growth and stemness maintenance.
