Polygalae Radix extract is a natural herbal preparation derived from the dried roots of Polygala tenuifolia or Polygala sibirica. It has been used for centuries in traditional Chinese medicine for its neuroprotective, memory-enhancing, antidepressant, anxiolytic, anti-inflammatory, and sedative properties[1][2][3][4]. The main active constituents include saponins (such as tenuifolin and polygalasaponins), oligosaccharide esters (like 3,6′-disinapoyl sucrose), xanthones, flavonoids, alkaloids and polysaccharides[1][2][3][9]. Mechanistically, preclinical studies suggest that the extract modulates glutamatergic synapses—particularly by influencing AMPA receptor activity—and increases brain-derived neurotrophic factor (BDNF) expression in the hippocampus[5]. It also demonstrates protective effects against amyloid-beta-induced neurotoxicity and axonal degeneration in models of Alzheimer’s disease[4], as well as rapid-onset antidepressant-like effects through modulation of glutamatergic neurotransmission[5]. Traditionally indicated for insomnia, forgetfulness/memory loss (including amnesia), depression/anxiety disorders and cough/palpitations; modern research supports its use primarily for cognitive impairment/neurodegenerative conditions and mood disorders. The raw form can cause gastrointestinal irritation due to saponins; thus it is often processed or combined with other herbs to reduce toxicity[1].
