Overview

Polygonatum odoratum polysaccharide 1 (POP1) is a neutral polysaccharide isolated from the rhizome of *Polygonatum odoratum (Mill.) Druce*, a plant widely used in traditional Chinese medicine for its yin-nourishing and hypoglycemic properties. POP1 is primarily composed of glucose and mannose residues and is characterized by specific structural features, including β-fructofuranose residues and 1→6 glycosidic bonds, which are essential for its biological activity. Mechanistically, POP1 ameliorates hyperglycemia by activating the phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt) signaling pathway. This activation leads to the downstream regulation of glycogen synthase kinase 3β (GSK3β) and Forkhead Box Protein O1 (FoxO1), resulting in enhanced hepatic glycogen synthesis and suppressed gluconeogenesis. Currently in preclinical development, POP1 is being investigated as a potential functional food or pharmaceutical supplement for the management of diabetes and hyperglycemia.
