Overview

Polygonatum sibiricum polysaccharide (PSP) is a collection of bioactive polysaccharides extracted from the rhizome of Polygonatum sibiricum, a traditional Chinese medicinal and food plant. PSPs are primarily composed of pectins, fructans, and glucogalactomannans, and exhibit various molecular weights depending on extraction and fractionation methods[1][2][4][8]. PSPs are characterized structurally as glucofructans and are not degraded in upper gastrointestinal digestion, reaching the colon where they serve as prebiotics for gut microbiota[8]. PSPs have diverse pharmacological activities, including immunomodulatory, antioxidant, anti-inflammatory, anti-fibrotic, anti-aging, anti-cancer, hypoglycemic, and lipid-lowering effects[1][3][5][6][7][9]. Mechanistically, PSPs can regulate immune cell activation and cytokine production (IL-2, TNF-α), exhibit anti-oxidative effects (e.g., upregulation of Klotho, regulation of calcium-phosphorus metabolism), inhibit inflammatory pathways, and modulate the TGF-β/Smad signaling pathway in liver fibrosis models[3][4][5][7]. Notably, their gut effects are partly mediated by restoration of the intestinal mucosal barrier and modulation of short-chain fatty acid production[7][8].
