Drug intelligence / Profile preview
PolyIC-LNPs are lipid nanoparticles encapsulating polyinosinic-polycytidylic acid (poly(I:C)), a synthetic analogue of double-stranded RNA that acts as an immune adjuvant by activating Toll-like receptor 3 (TLR3). The lipid nanoparticle (LNP) formulation is designed to enhance intracellular delivery of poly(I:C) to target cells, protect the payload from degradation, and improve cellular uptake and endosomal escape. PolyIC-LNPs are being studied primarily as immunostimulatory agents or vaccine adjuvants for cancer immunotherapy, infectious disease vaccines, and other applications requiring robust activation of the innate immune response[6][5]. The LNP delivery system typically contains an ionizable cationic lipid, helper phospholipid, cholesterol, and PEGylated lipid, and is designed for optimal loading, stability, and biodistribution of the poly(I:C) payload[9][5].
