Polymerized Alternaria alternata allergen extract is a chemically modified allergen extract (allergoid) derived from the fungus Alternaria alternata, intended for use in allergen-specific immunotherapy. The extract is prepared by depigmenting, then chemically polymerizing native Alternaria allergens with glutaraldehyde, resulting in high-molecular-mass complexes with reduced IgE-binding activity while retaining immunogenicity. Major allergens in the polymer include Alt a 1, Alt a 3, Alt a 6, and Alt a 8. This modification aims to reduce the risk of adverse reactions (notably, less IgE-mediated reactivity) compared to native extracts, while still promoting the induction of protective IgG antibodies capable of blocking allergen–IgE interactions. Polymerized extracts demonstrate strong potential for safer and effective immunotherapy in both adults and children with allergies to Alternaria alternata, and are especially suitable for those with polysensitization, as the reduced enzymatic/proteolytic activity decreases degradation of other combined allergens in mixed formulations. The immunological mechanism chiefly involves immune modulation (regulatory T cell and Th1 response, increased IL-10, suppressed Th2 reactivity), resulting in increased tolerance to Alternaria antigens.
