Polyoxyethanyl alpha-tocopheryl sebacate (PTS) is a synthetic, water-soluble derivative of alpha-tocopherol (Vitamin E) that functions as an amphiphilic surfactant and drug delivery vehicle. It is primarily utilized to solubilize highly lipophilic molecules, most notably Coenzyme Q10 (CoQ10), by forming stable, water-soluble non-covalent complexes, typically at a 2:1 molar ratio (PTS:CoQ10). Following oral administration, PTS is hydrolyzed by intestinal esterases to release free alpha-tocopherol, which acts as a potent antioxidant. Preclinical research, largely conducted by the National Research Council of Canada, has investigated PTS-CoQ10 formulations for neuroprotection in models of ischemic stroke, cardiac arrest, and traumatic brain injury, where it facilitates the delivery of antioxidants across biological membranes to mitigate oxidative damage.
