Overview

Polyphyllin VI is a natural steroidal saponin isolated from the rhizomes of *Paris polyphylla*, a plant long used in traditional Chinese medicine for its anti-tumor properties. It has demonstrated significant anti-cancer activity, particularly in colorectal cancer models such as DLD-1 cells. The mechanism of action for polyphyllin VI involves the induction of autophagy-mediated cell death, characterized by the upregulation of autophagy markers (such as LC3-II). Notably, this process appears to be independent of the p53 and caspase-3 apoptotic pathways. Furthermore, polyphyllin VI has shown potential as a chemosensitizer, enhancing the cytotoxic effects of conventional chemotherapeutic agents like doxorubicin, which suggests its potential utility as a complementary therapy for advanced or recurrent colorectal carcinoma.
