Overview

Polyporus polysaccharide (PPS) is a bioactive compound extracted from the medicinal fungus Polyporus umbellatus and related species. It is primarily composed of high-molecular-weight polysaccharides and has demonstrated a range of pharmacological activities including immunomodulatory, anticancer, antioxidant, anti-inflammatory, antifibrotic, and hepatoprotective effects. Mechanistically, PPS exerts its effects by modulating immune responses (stimulating natural killer cells, T cells, B cells, and macrophages), inhibiting tumor cell proliferation (notably in bladder cancer), suppressing myofibroblast differentiation via inhibition of the TGF-β/Smad2/3 pathway in pulmonary fibrosis models[1][5], inducing autophagy in cancer cells through macrophage regulation[2][6], and enhancing cytokine production such as NO, IL-6 and TNF-α by activated macrophages[5]. In China, both capsule and injectable forms have been approved for clinical use—most notably for hepatitis B treatment—and are also used as adjuncts in cancer therapy[4].
