Polysaccharide-berberine is an experimental, colon-targeted hydrogel therapeutic engineered for the treatment of ulcerative colitis. It consists of rhubarb-derived polysaccharides and berberine-loaded dendrimers that self-assemble via intermolecular hydrogen bonding and electrostatic interactions. This formulation is designed for oral administration, allowing for localized accumulation in colonic tissues. Its therapeutic effects are mediated by promoting the polarization of anti-inflammatory M2 macrophages, suppressing the secretion of pro-inflammatory cytokines, and remodeling the dysbiotic gut microbiome and metabolic environment to restore intestinal homeostasis.
