Polysialic acid is a naturally occurring polymer of N-acetylneuraminic acid with remarkable biological properties. ## Description Polysialic acid is a highly hydrophilic, biodegradable polymer of N-acetylneuraminic acid (sialic acid) with restricted expression in the body. It has no known receptors in the body and exhibits long half-lives in blood circulation after intravenous injection[1]. PolySia is best known for its role in modulating neuronal development, but it also has significant functions in the immune system[2][3]. The polymer has several important biological properties: 1. It provides protection against proteolytic enzymes for conjugated proteins In the immune system, polySia expression is dynamically regulated during differentiation, maturation, and activation of various immune cells, including NK cells, T cells, macrophages, and hematopoietic progenitors[2][5][8]. ## Therapeutic Applications PolySia has several potential therapeutic applications: 1. **Drug delivery carrier**: Due to its long half-life in circulation, polySia has been proposed as a carrier for short-lived drugs and small peptides[1]. 2. **Protein stabilization**: Covalent coupling of polySia to enzymes like catalase and asparaginase significantly increases their stability against proteolytic degradation and extends their half-life in vivo[1]. 3. **Anti-inflammatory agent**: PolySia has shown neuroprotective properties in models of Parkinson's disease by preventing exacerbated immune responses[3]. 4. **Histone neutralization**: PolySia in human plasma can bind histones and reduce histone-mediated cytotoxicity[9]. 5. **Alternative to PEG**: PolySia serves as a biodegradable alternative to non-biodegradable monomethoxypoly(ethylene glycol) for increasing protein half-life[1].
