Overview

PolySTING is a polymeric STING (Stimulator of Interferon Genes) agonist developed to enhance antitumor immune responses. It is designed to activate the STING pathway, primarily through the licensing of type I conventional dendritic cells (cDC1), which triggers myeloid activation and the production of type I interferons. This mechanism facilitates the recruitment and activation of cytotoxic T cells, potentially overcoming resistance to immune checkpoint inhibitors. Developed by researchers at UT Southwestern Medical Center, PolySTING has demonstrated efficacy in preclinical models and is being evaluated using ex vivo patient-derived tumor fragment (PDTF) platforms to identify functional biomarkers for clinical outcomes, such as post-surgical recurrence in head and neck squamous cell carcinoma (HNSCC).
