Polyvalent antigen-KLH conjugate vaccine is a multivalent cancer vaccine comprised of five tumor-associated antigens (TAAs): Globo H, GM2 ganglioside, Tn-MUC1, TF, and sTn conjugated with the immunoadjuvant keyhole limpet hemocyanin (KLH). The vaccine is designed to induce antibody responses against these tumor-associated antigens. KLH serves as a hapten carrier and immunostimulant to improve immune recognition of the antigens. The conjugation to KLH is achieved through various methods including covalent attachment using m-maleimidobenzoyl-N-hydroxysuccinimide ester (MBS) for TF(c) and Tn-MUC1, MMCCH linker for Globo H, and reductive amination methods for GM2. The vaccine is typically administered with an immunological adjuvant such as OPT-821 (derived from Quillaja saponaria Molina).
