Polyvalent vaccine-KLH conjugate + OPT-821 is a multivalent cancer immunotherapy developed by Memorial Sloan Kettering Cancer Center (MSK). It consists of four tumor-associated carbohydrate antigens (TACAs)—GM2, Globo-H, Tn-MUC1, and TF(c)—each individually conjugated to the carrier protein keyhole limpet hemocyanin (KLH) to overcome self-tolerance and enhance immunogenicity. The formulation includes OPT-821, a purified saponin-based adjuvant (QS-21), which further stimulates the immune response. The vaccine is designed to induce high-titer IgM and IgG antibodies against antigens frequently overexpressed on the surface of epithelial ovarian, fallopian tube, and primary peritoneal cancer cells. It has been primarily evaluated in the setting of clinical remission to delay or prevent recurrence, often in combination with the anti-VEGF monoclonal antibody bevacizumab.
