Polyzincliothyronine is an investigational, extended‑release formulation of liothyronine (L‑triiodothyronine, T3) created by coordinating T3 with zinc to form a supramolecular polymeric complex that adheres to the intestinal mucosa and slowly releases active hormone over time.[1][4][5][7] The complex consists of divalent zinc ions bound to tridentate T3 ligands, producing a mucoadhesive depot in the small intestine from which T3 is liberated by ligand exchange and absorbed, blunting the high peak concentrations and rapid clearance seen with conventional liothyronine while maintaining T3‑dependent biological effects.[1][4][5] Developed by Synthonics in collaboration with academic investigators, polyzincliothyronine is being studied primarily for hypothyroidism and related conditions where more physiologic, stable T3 exposure is desired compared with immediate‑release liothyronine.[1][5][7]
