pomalidomide + obinutuzumab

This investigational combination therapy, referred to as the GP regimen, consists of pomalidomide and obinutuzumab. Pomalidomide is a third-generation immunomodulatory drug (IMiD) that acts as a small molecule molecular glue, binding to the cereblon (CRBN) E3 ubiquitin ligase complex to induce the degradation of lymphoid transcription factors, resulting in antiproliferative and immunomodulatory effects. Obinutuzumab is a glycoengineered, humanized type II anti-CD20 monoclonal antibody that targets CD20 on B-cells, inducing direct cell death and enhancing antibody-dependent cellular cytotoxicity (ADCC). This chemotherapy-free approach is being evaluated by the Institute of Hematology & Blood Diseases Hospital in China for the treatment of relapsed or refractory indolent lymphomas, including follicular lymphoma, marginal zone lymphoma, and chronic lymphocytic leukemia/small lymphocytic lymphoma.
