POMP siRNA is a small interfering RNA (siRNA) therapeutic candidate designed to silence the expression of the proteasome maturation protein (POMP). POMP is a molecular chaperone essential for the assembly of the 20S proteasome core. By degrading POMP mRNA through the RNA interference (RNAi) pathway, the drug impairs proteasome biogenesis and activity. This leads to the accumulation of pro-apoptotic proteins and tumor suppressors, such as p53 and cyclin-dependent kinase inhibitors, resulting in cell cycle arrest and apoptosis in tumor cells. Research indicates that suppressing POMP can overcome resistance to proteasome inhibitors like bortezomib, particularly in ERα-positive breast cancers. This mechanism mimics the endogenous regulatory role of microRNA-101 (miR-101), which naturally targets POMP to control protein turnover and tumor cell proliferation.
