Ponatinib + cytarabine is a combination therapy used primarily in hematological malignancies. Ponatinib is a multi-target tyrosine kinase inhibitor that targets BCR-ABL including the T315I mutation, as well as VEGFR, PDGFR, FGFR, EPH receptors, KIT, RET, TIE2, and FLT3. Cytarabine is a cytotoxic antimetabolite that inhibits DNA synthesis. This combination has been studied in blast-phase chronic myeloid leukemia, FLT3-mutated acute myeloid leukemia, and Philadelphia chromosome-positive acute lymphoblastic leukemia.
