Overview

Pontmestrocel is an allogeneic cell therapy product composed of human umbilical cord-derived mesenchymal stem cells (hUC-MSCs). Developed by Tianjin Madison Regenerative Medicine, it is primarily investigated for the treatment of chronic graft-versus-host disease (cGVHD), a debilitating complication of allogeneic hematopoietic stem cell transplantation. The therapeutic mechanism of pontmestrocel relies on the innate immunomodulatory and anti-inflammatory properties of mesenchymal stem cells. These cells secrete various paracrine factors and interact directly with immune cells to suppress the pathological activation of T cells and B cells, promote the expansion of regulatory T cells (Tregs), and modulate dendritic cell function. By restoring immune homeostasis, pontmestrocel aims to alleviate the multi-organ inflammation and fibrosis characteristic of cGVHD. It is currently undergoing clinical evaluation in China, specifically targeting patients with steroid-refractory or steroid-dependent forms of the disease.
