Overview

Porcine anti-human lymphocyte immunoglobulin (p-ATG or pALG) is a polyclonal antibody preparation derived from pigs immunized with human thymocytes or peripheral blood lymphocytes. It is primarily composed of IgG and functions as an immunosuppressive agent by targeting and depleting human T cells, leading to transient suppression of the immune response. The main mechanism involves binding to various surface antigens on T cells, resulting in their depletion through apoptosis, complement-dependent cytotoxicity (CDC), and antibody-dependent cellular phagocytosis (ADCP). This drug is used mainly for the prevention and treatment of graft-versus-host disease (GVHD) following hematopoietic stem cell transplantation, as well as for first-line immunosuppressive therapy in severe aplastic anemia—especially in China where it has been widely adopted since 2009. It has also been investigated for induction therapy in kidney transplantation[1][4][6][8].
