Porphyrin high-density lipoprotein nanoparticles (Por-HDL-NPs), also known as PLP, are biomimetic theranostic nanoparticles engineered for applications in oncology and cardiovascular disease. These nanoparticles consist of porphyrin-lipid conjugates integrated into a high-density lipoprotein (HDL) framework, often utilizing apolipoprotein A-I (ApoA-I) or mimetic peptides like reverse-4F (R4F). In the context of atherosclerosis, they target scavenger receptor class B type 1 (SR-B1) on macrophages, enabling near-infrared fluorescence (NIRF) and positron emission tomography (PET) imaging while exerting anti-inflammatory effects and promoting cholesterol efflux. In cancer treatment, their small size (~20 nm) facilitates deep tumor penetration and cellular uptake, where the porphyrin component serves as a photosensitizer for photodynamic therapy (PDT). This process induces local tumor ablation and stimulates systemic antitumor immunity.
