Overview

PORT-6 + PORT-7 + pembrolizumab is a combination immunotherapy regimen being developed by Portage Biotech in collaboration with Merck. The combination consists of two small molecule adenosine receptor antagonists—PORT-6 (formerly TT-10), which targets the adenosine 2A receptor (A2AR), and PORT-7 (formerly TT-4), which targets the adenosine 2B receptor (A2BR)—alongside the anti-PD-1 monoclonal antibody pembrolizumab (Keytruda). This therapeutic strategy aims to overcome adenosine-mediated immunosuppression within the tumor microenvironment, thereby enhancing the anti-tumor activity of PD-1 checkpoint blockade. The combination is currently being evaluated in the ADPORT-601 Phase 1a/1b clinical trial, specifically targeting patients with 'adenosine-high' solid tumors, including non-small cell lung, prostate, renal, head and neck, colorectal, endometrial, and ovarian cancers.
