PORT-7 is a small molecule, oral adenosine A2B-receptor (A2BR) antagonist being developed by Portage Biotech. It selectively inhibits the adenosine A2B receptor, which plays a role in immunosuppression within the tumor microenvironment. Preclinical studies have shown promising efficacy in mesothelioma models, demonstrating superior single-agent activity compared to anti-PD1 antibody treatment. When combined with anti-PD1 antibody, PORT-7 showed enhanced anti-tumor effects with over 90% tumor growth inhibition in murine mesothelioma models. The combination therapy also led to the formation of tertiary lymphoid structures and increased immune effector cells in tumors, indicating a favorable immune response.
