Porustobart is a fully human anti-CTLA-4 monoclonal heavy chain only antibody (HCAb) developed by Harbour BioMed using their proprietary Harbour Mice platform. It is the first heavy chain only fully human antibody to enter clinical development globally. Porustobart works through two main mechanisms of action: inhibition of negative signaling from the interaction of CTLA-4 with the co-stimulatory molecule B7 and depletion of immunosuppressive regulatory T cells (Treg) in tumor tissues via enhanced antibody-dependent cell cytotoxicity (ADCC). This dual mechanism leads to potent anti-tumor efficacy and a differentiated pharmacokinetic profile with durable pharmacodynamic effects. Porustobart has demonstrated promising activity and manageable safety in early-phase trials for multiple solid tumors including melanoma, hepatocellular carcinoma (HCC), neuroendocrine tumors (NET/NEC), renal cell carcinoma, colorectal cancer, and other solid tumors[1][2][5].
