Overview

Porustobart + tislelizumab is a combination of two monoclonal antibodies used in oncology immunotherapy. **Porustobart** (HBM4003) is a fully human heavy chain only monoclonal antibody targeting CTLA-4, developed using Harbour Mice technology by Harbour BioMed. **Tislelizumab** (BGB-A317) is a humanized IgG4 anti–PD-1 monoclonal antibody from BeiGene, designed to minimize Fcγ receptor binding on macrophages to reduce antibody-dependent phagocytosis, a known resistance mechanism to anti–PD-1 therapy. The combination aims to synergistically enhance immune-mediated tumor cell killing by jointly inhibiting negative immune checkpoints CTLA-4 and PD-1, respectively. It's being investigated in phase I/II studies for advanced solid tumors, such as melanoma and metastatic colorectal cancer. Early data suggest promising anti-tumor activity and a manageable safety profile in immunotherapy-naive patients, including those with difficult-to-treat subtypes like mucosal melanoma[1][5].
