Poseltinib + lenalidomide + glofitamab (also known as the GPL regimen or Project 201) is an investigational chemotherapy-free combination therapy being developed by NOBO Medicine for the treatment of relapsed or refractory diffuse large B-cell lymphoma (R/R DLBCL). The regimen combines poseltinib, an orally active, irreversible Bruton's tyrosine kinase (BTK) and TEC kinase inhibitor; lenalidomide, an immunomodulatory drug (IMiD) that binds to cereblon (CRBN) to promote the proteasomal degradation of transcription factors IKZF1 and IKZF3; and glofitamab, a CD20xCD3 T-cell engaging bispecific monoclonal antibody. This combination leverages the synergistic effects of BTK inhibition, immunomodulation, and T-cell-mediated cytotoxicity to target and eliminate malignant B cells. The regimen is currently being evaluated in a Phase II clinical trial.
